Circular RNA circDLC1 inhibits MMP1-mediated liver cancer progression via interaction with HuR

Rationale: circular RNAs (circRNAs) have been demonstrated to play a crucial role in cancer progression. KIAA1429, key component of the m6A methyltransferase complex, has recently reported promote hepatocellular carcinoma (HCC) progression by regulating methylation. The aim present study is investigate KIAA1429-mediated HCC Methods: RNA sequencing (RNA-seq) and methylated immunoprecipitation (m6A-seq) were utilized identify KIAA1429-regulated circRNAs. effects circDLC1 on proliferation metastasis hepatoma cells examined vitro vivo. RT-qPCR was used measure expression tissues cells. FISH, RIP assays biotin-labeled pull-down downstream effector circDLC1. targets identified using RNA-seq. Results: Our data that downregulated closely relevant favorable prognosis. Overexpression inhibited motility vivo, while silencing played opposite role. Mechanistic investigations revealed could bind RNA-binding protein HuR, which subsequently reduced interaction between HuR MMP1 mRNAs, thus MMP1, ultimately contributing inhibition Conclusion: work suggests circDLC1, target promising prognostic marker for patients, circDLC1-HuR-MMP1 axis may serve as potential therapeutic treatment.
